Yeast
surface display (YSD) is a powerful protein engineering technology and
an important high-throughput screening platform in protein engineering.
Yeast cells have eukaryotic post-translational modification mechanisms
that can help target proteins fold correctly and can be used to display
various eukaryotic proteins, including antibodies, receptors, enzymes,
and antigenic peptides.
Technical Principle
The
AlpVHHs yeast surface display platform uses the "α-lectin" display
system to insert the nanobody gene sequence and the expression tag HA
tag gene sequence into the display plasmid vector of the protein
scaffold-Aga2 gene. The vector uses nutritional markers to maintain
selective growth in yeast, and galactose is added to the culture medium
to induce yeast to display on the surface. The nanobody fusion protein
is secreted and anchored on the yeast cell wall. Combined with magnetic
beads and flow sorting technology for multiple rounds of screening,
high-affinity and high-stability clones can be screened.
