Nanobodies
(Nbs) are versatile molecular tools that are used in many areas of
biological sciences, such as immunofluorescence microscopy and
structural biology. In order to meet detection needs, nanobodies often
need to be labeled.
A
large amount of data statistics show that more than half of the
interactions are mediated by CDR3, CDR2 accounts for 20%, and CDR1
accounts for 13%. Surprisingly, 16% of the interactions are established
by residues in the nanobody framework (non-CDR), which are concentrated
in four structurally different regions, and non-CDR interactions are
mainly mediated by residues in the framework 2 region, and the residues
involved are mostly charged residues. Nanobodies have a relatively
delicate structure, and conventional active amino-based labeling can
easily inactivate antibodies. At the same time, this type of labeling
method is difficult to control the labeling ratio. Critical Point
specializes in nanobody development and can provide fixed-point nanobody
labeling services, which can not only maintain antibody activity but
also clarify the labeling ratio.

II. Service Process
1.
The customer submits a request for commissioned marking and specifies
the marking requirements, agrees with the company on specific
experimental items, quantity and costs, and both parties sign the
"Commissioned Marking Preparation Contract".
2.
The customer provides samples to be marked, and our company arranges
marking services in accordance with the contract requirements and
completes the marking commission within the specified time.
3.
After the marking is completed, our company will package according to
the customer's requirements, mail them to the customer free of charge at
low temperature, and provide a marking report at the same time.
Ⅲ. Cases

AlpVHHs uses
optimized site-specific coupling technology to ensure that antibodies
are efficiently labeled with biotin while ensuring that antibody
affinity is not affected, facilitating subsequent testing and
application.

AlpVHHs uses
optimized fluorescent dye labeling technology to test and optimize the
coupling with a variety of fluorescent dyes to determine the best
antibody: dye molecule ratio, thereby producing a conjugate with the
highest specific signal intensity. At the same time, multiple
technologies are used to optimize bioconjugation chemistry and strict
purification to remove free dyes and antibodies, thereby producing
higher quality products.
