Anti-VHH, AlpHcAbs® Rabbit Antibody (HRP) is a high-affinity anti VHH antibody developed for the specific detection of camelid single-domain antibodies (VHH), commonly known as nanobodies. This HRP-conjugated reagent serves as a reliable and highly sensitive tool for nanobody detection in ELISA and Western blot, delivering excellent specificity with minimal background in recombinant antibody studies and phage display workflows.
Anti-VHH, AlpHcAbs® Rabbit Antibody (HRP) is a recombinant anti-camelid VHH antibody designed for the specific recognition of nanobody VHH domains from alpaca, llama, and camel. It features high affinity, superior sensitivity, and excellent lot-to-lot consistency, enabling reliable and reproducible single domain antibody detection. Validated by immunoelectrophoresis and ELISA, this high specificity anti-VHH antibody for nanobody detection selectively binds camelid VHH while showing no cross-reactivity with conventional IgG from mouse, rat, rabbit, goat, or human, making it ideal for nanobody expression analysis, purification monitoring, and immunoassay development.

Key Features and Advantages
High specificity for VHH domains
Selectively recognizes camelid VHH fragments without cross-reactivity to conventional IgG heavy and light chain structures.
HRP conjugation for signal amplification
Horseradish peroxidase (HRP) labeling enables sensitive detection in enzyme-based assays such as ELISA and Western blot.
Recombinant production for consistency
Eliminates variability associated with traditional animal-derived secondary antibodies.
Low background and high signal-to-noise ratio
Suitable for assays requiring high sensitivity and minimal nonspecific binding.
Background:
The Camelidae family includes camels, llamas, alpacas, and vicuñas. These species produce two distinct types of IgG in their serum: conventional heterotetrameric IgG1 antibodies and unique homodimeric heavy-chain antibodies (HCAbs) known as IgG2 and IgG3.
Unlike conventional IgG1, HCAbs lack light chains. The smallest functional antigen-binding unit of HCAbs is the single-domain VHH fragment, commonly called a nanobody (Nb). Derived from the variable region of heavy chains, VHH antibodies are extremely small (<15 kDa), offering superior tissue penetration and making them highly valuable for research, diagnostics, and therapeutic applications.
Applications
This anti-VHH secondary antibody is suitable for a range of immunodetection workflows involving nanobody-based reagents:
·Nanobody detection in ELISA assays
·Western blot analysis of recombinant VHH antibodies
·Detection of camelid nanobodies in recombinant antibody research
·Monitoring nanobody expression and purification
·Screening VHH antibodies from phage display libraries
·Detection of nanobody-tagged proteins
Performance:
ELISA: 1:4000-1:10000
WB: 1:4000-1:10000
Dilution factors are presented in the form of a range because the optimal dilution is a function of many factors, such as antigen density, permeability, etc. The actual dilution used must be determined empirically.
