In the development of nanobodies, Freund's adjuvant, commonly used, is highly toxic. Injection sites can easily cause symptoms such as calluses, suppuration, and even severe arthritis, making it very unfriendly to animals. Secondly, Freund's adjuvant is a water-in-oil adjuvant, requiring emulsification of the antigen during immunization, disrupting its conformation, which is very cumbersome. Extensive testing of immune sera revealed that Freund's adjuvant primarily produces IgG1, with a low proportion of single-domain antibodies. Case Study:
