Development of anti small molecule antibodies

Small molecule compounds refer to pesticides, veterinary drugs, biological toxins, environmental pollutants, hormones, heavy metals and other substances with a molecular weight of no more than 1000 Daltons. Trace detection of small molecule compounds is of great significance in agriculture, food safety, environmental monitoring, medical diagnosis and other fields. Immunoassay technology based on antigen/antibody specific recognition has the advantages of strong specificity, high sensitivity, good stability, simple and fast operation.


The key to the preparation of antibodies for small molecule compounds is hapten design. Since small molecule compounds have relatively small molecular weights and only have reactive properties but no immunogenicity, small molecule haptens must be coupled with macromolecular carrier proteins in order to prepare specific antibodies against haptens through various antibody technologies.


Common small molecule compound antibody preparations are mainly rabbit polyclonal antibodies and mouse hybridoma monoclonal antibodies. Rabbit polyclonal antibodies have multiple sources and simple methods for preparation, and were widely used in the early stage, but their specificity is not high and the batch differences are large, which limits the specificity and repeatability of immunoassay methods. Mouse monoclonal antibodies have the advantages of strong specificity and easy batch production, but the whole process involves more materials and operation techniques, and each step may affect the quality of the final antibody.


Based on the HTS-Smart Select platform, AlpVHHs uses immunization with small molecule complex antibodies to allow animals to produce anti-isotypic antibodies, and then constructs a yeast display library. Through flow cytometry analysis and sorting, it can finely control the screening process and efficiently screen high-affinity and high-specificity antibodies.
