The core application of phage display technology is antibody display. Currently, different phage systems are available for phage display, such as T4, λ, and filamentous M13 phage. These different phage systems each have their advantages and disadvantages, with M13 phage being the most widely used. M13 enzyme-labeled antibodies are essential reagents for ELISA assays used to screen results using phage display technology.
Introduction of M13 phage
M13 phage belongs to the filamentous phage family, collectively known as Ff phage. Ff phage only infects Escherichia coli strains expressing F pili, as phage attachment to bacteria requires the binding of the phage coat protein to the tips of the F pili. M13 phage is neither temperate nor lytic. Instead, it establishes a chronic infection in the host, continuously releasing new phage. M13 phage contains a 6407-base single-stranded DNA (ssDNA) genome, comprised of nine genes encoding 11 different proteins (five of which are coat proteins, while the remaining six are involved in phage replication and assembly).
