I. Phage Display

In October 2018, half of the Nobel Prize in Chemistry was awarded to American scientist George P. Smith and British scientist Gregory P. Winter for their development of phage display technology for peptides and antibodies. Phage display technology has extensive applications in antibody discovery and biopharmaceutical research, serving as a foundational technology for the development of new biological drugs. II. M13 Phage

The most commonly used and established phage display system is based on M13. M13 is a filamentous phage composed of a circular single-stranded DNA (ssDNA) over 6,400 nucleotides long, encapsidated by approximately 2,700 copies of the major capsid protein P8, terminated by five copies of two different minor capsid proteins (P9, P6, and P3). Infection with this filamentous phage is not lethal; however, it can cause turbid plaques in Escherichia coli. It is a non-lytic virus. However, a decrease in cell growth can be observed in infected cells. M13 plasmids are used in many recombinant DNA processes, and the virus is also being studied for applications in nanostructures and nanotechnology. These features make M13 an indispensable platform for modern antibody discovery research.
